Are you studying signal transduction? Would you like to skip the Western blots and get a faster, more direct readout of what’s happening in your cultured cells?

First described in 2002, the In Cell Western assay was developed by LI-COR scientists looking for a better way to quantify proteins in cultured cells. Analyzing many samples or time points by Western blot is labor-intensive and time-consuming, and introduces the potential for error. In contrast, In Cell Western cell-based assays allow you to grow your cells in 96- or 384-well plates, treat them with the appropriate conditions, and then quickly fix them to capture a snapshot of protein expression and phosphorylation levels at that moment in time. Cells are then stained with primary and secondary antibodies against proteins of interest, in a process that is conceptually similar to immunofluorescent staining for microscopy. After staining, the total fluorescent signals from each well are imaged and quantified, allowing you to make ensemble measurements of the cell populations in each well. The ability to stain and image simultaneously in two fluorescence channels allows you to improve accuracy by normalizing for well-to-well variation in cell number – or, if you prefer, stain with antibodies against two different targets to maximize the data output from each plate. The assay format is particularly well suited to phosphoprotein and signal transduction analysis.
